The traditional form 間 stacks 日 (sun) inside 門 (door). Picture sun squeezing through the narrow crack of a doorway. That gap — the space between two things — is the original meaning. Later it stretched to mean a room (the space between walls) and a stretch of time (the space between moments).
You can't count time with 个. For a stretch of time, use 一段时间 (yí duàn shíjiān).
中间 already means 'the middle of.' Adding 里 makes it sound like 'inside the middle.' Just say 房间中间.
Nope. 间 never stands alone — 'room' is always 房间.
